Overview

Dermatophagoides farinae major allergen Der f 1 is a 25-27 kDa cysteine protease and a primary allergen produced by the American house dust mite (UniProt P08176). It is predominantly found in mite fecal pellets and plays a critical role in the pathogenesis of allergic diseases such as asthma, rhinitis, and atopic dermatitis (Allergen.org). The protein's enzymatic activity facilitates allergen sensitization by disrupting epithelial tight junctions and cleaving cell surface receptors like CD23 and CD25, which promotes a Th2-biased immune response (PubMed: 15707969). Der f 1 is a central component in allergen-specific immunotherapy (AIT), where controlled exposure to the allergen aims to induce desensitization and long-term immunological tolerance (NIH: PMC7465134). Therapeutic strategies involving Der f 1 focus on modulating the IgE-mediated allergic cascade, often by shifting the immune profile toward a Th1/Treg response and increasing protective IgG4 antibodies (ClinicalTrials.eu).

Mechanism of action

Allergen-specific immunotherapy (AIT) induces immunological tolerance by shifting the immune response from Th2 to Th1/Treg, increasing IL-10 and TGF-beta, and promoting allergen-specific IgG4 antibodies that block IgE-mediated activation (NIH: PMC7465134).
