Dermatophagoides siboney allergens are a complex group of proteins produced by the tropical house dust mite species D. siboney, which is a major source of indoor allergens in the Caribbean and surrounding regions (Ferrándiz et al., 1996, PMID: 8646100). The most clinically significant components are Der s 1, a cysteine protease, and Der s 2, a member of the NPC2-like lipid-binding protein family (WHO/IUIS Allergen Nomenclature, allergen.org). These allergens act as potent antigens that trigger Type I hypersensitivity reactions by cross-linking IgE on the surface of mast cells and basophils, leading to the release of inflammatory mediators (PubMed: 15985814). In the context of therapeutic intervention, these allergens are the primary components of allergen-specific immunotherapy (AIT) formulations, such as Valergen-DS (Labarta et al., 2006, PMID: 16846561). The goal of such therapy is to desensitize the patient's immune system, promoting the development of regulatory T cells and the production of protective IgG4 antibodies. Understanding the molecular structure and biological activity of these allergens is essential for improving diagnostic accuracy and the efficacy of immunotherapy for allergic asthma and rhinitis (UniProt: P49274, P49275).
