Dermatophagoides siboney allergens are a complex group of proteins produced by the house dust mite species D. siboney, which is a major source of indoor allergens in tropical and subtropical regions, particularly Cuba (Ferrándiz et al., 1996, PMID: 8952544). These allergens include several well-characterized proteins such as Der s 1 (a cysteine protease), Der s 2 (a lipid-binding protein of the NPC2 family), and Der s 3 (a serine protease), which are officially recognized by the WHO/IUIS Allergen Nomenclature Sub-Committee. In sensitized individuals, these proteins act as antigens that cross-link IgE on the surface of mast cells and basophils, triggering the release of inflammatory mediators that lead to allergic asthma, rhinitis, and atopic dermatitis. From a therapeutic perspective, these allergens are the primary components of allergen-specific immunotherapy (AIT) products, such as the VALERGEN-DS extract, which are designed to desensitize the immune system through controlled exposure (Borroto et al., 2003, PMID: 12846871). Understanding the molecular composition of these allergens is essential for the development of standardized diagnostic tools and effective vaccines tailored to regional mite populations.
Allergen-specific immunotherapy (AIT) utilizes these allergens to induce immunological tolerance by shifting the immune response from a Th2-dominated profile to a Th1/Treg profile, increasing the production of blocking antibodies such as IgG4 and reducing IgE-mediated mast cell degranulation (Borroto et al., 2003, PMID: 12846871).
