Dermo-epidermal tissue replacements are engineered constructs that mimic the structure and function of natural human skin by incorporating both a dermal layer (typically containing fibroblasts within an extracellular matrix) and an epidermal layer (typically composed of keratinocytes). The goal is to restore full-thickness skin defects with functional, vascularized tissue that supports re-epithelialization and integration with host tissues. They rely on scaffold biocompatibility/porosity for cell migration/nutrient diffusion, proper formation/mimicry of the basement membrane zone (DEJ), and paracrine signaling between keratinocytes/fibroblasts/endothelial cells.
