Overview

Desmoglein-1 (DSG1) is a calcium-dependent transmembrane glycoprotein belonging to the cadherin superfamily, specifically the desmosomal cadherins[1][3][6]. DSG1 is predominantly expressed in the upper layers of stratified epithelia, especially the epidermis, where it mediates strong cell-cell adhesion as a critical component of desmosomes, helping to maintain tissue cohesion and barrier function[1][2][3]. In addition to its structural adhesive role, DSG1 participates in intracellular signaling pathways such as EGFR/Erk1/2, promoting keratinocyte differentiation and suprabasal morphogenesis[2][5]. DSG1 dysfunction is associated with several diseases, most notably the autoimmune disorder pemphigus foliaceus, in which autoantibodies target DSG1 and disrupt skin integrity, as well as genetic disorders such as striate palmoplantar keratoderma and SAM syndrome[1][3][5][6]. Detection of anti-DSG1 antibodies serves as a key biomarker in diagnosis, and immunosuppressive therapies are utilized in management when DSG1 is the autoimmune target[1][3].
