Desmoglein 3 (DSG3) mRNA encodes a calcium-dependent transmembrane glycoprotein that is a critical component of desmosomes, the intercellular junctions responsible for maintaining the structural integrity of stratified squamous epithelia (UniProt: P32926). The protein product of this mRNA facilitates cell-cell adhesion by interacting with other desmogleins and desmocollins, effectively anchoring the intermediate filament network to the plasma membrane (NCBI Gene: 1830). DSG3 is clinically significant as the primary autoantigen in Pemphigus Vulgaris, where autoantibodies against the protein lead to the loss of cell adhesion and severe mucosal blistering (PubMed: 15542702). In oncology, DSG3 mRNA is frequently overexpressed in squamous cell carcinomas, particularly in the lung and head and neck, where it promotes tumor cell proliferation and migration (PubMed: 21602877). Furthermore, its high specificity for squamous cell lineages makes it a robust biomarker for differential diagnosis in pulmonary pathology (PubMed: 21602877). Consequently, DSG3 mRNA has emerged as a potential therapeutic target for RNA-based interventions, such as small interfering RNAs (siRNAs), which aim to downregulate its expression to inhibit tumor progression (PubMed: 25670454). Therapeutic development focusing on this mRNA must carefully balance efficacy against the risk of disrupting normal epithelial adhesion in healthy tissues.
