Developmental pluripotency associated 2 (DPPA2) is a small nuclear protein (~300 amino acids) expressed predominantly in pre-implantation embryos and germ cells. It acts as an epigenetic priming factor, binding to GC-rich promoter regions via its SAP domain and interacting with histones to regulate chromatin accessibility. DPPA2, usually as a heterodimer with DPPA4, maintains open chromatin states at developmental gene loci, preventing premature DNA methylation and facilitating proper differentiation. It is critical for zygotic genome activation and reprogramming somatic cells to pluripotency. DPPA2 expression is tightly regulated and silenced after gastrulation, and knockout studies show that its absence impairs differentiation efficiency and causes developmental defects, notably after birth. There are no approved drugs targeting DPPA2, but its role as a biomarker for pluripotency and links to cancer make it a significant molecule in stem cell biology and regenerative medicine research
