Overview

Developmental pluripotency-associated protein 3 (DPPA3) is a maternally expressed protein highly enriched in germ cells, pre-implantation embryos, and embryonic stem cells. It plays a critical role in epigenetic regulation, especially safeguarding DNA methylation landscapes in zygotes and oocytes, both by inhibiting maintenance DNA methylation and by modulating the interaction and subcellular localization of proteins like UHRF1 and DNMT1. DPPA3 is necessary for normal early embryonic development and maintenance of pluripotency, acting as a marker gene for naïve pluripotent stem cells. It is not a classic therapeutic target, but perturbations of DPPA3 or its pathways disrupt the stability of key pluripotency proteins (e.g., Nanog), leading to abnormal embryonic development and altered epigenetic programming.
