Overview

Developmentally regulated endothelial locus 1 (DEL-1), also known as EGF-like repeats and discoidin I-like domains 3 (EDIL3), is a secreted glycoprotein primarily produced by endothelial cells and certain macrophage subsets. It plays critical roles in vascular biology, immune regulation, inflammation resolution, and tissue remodeling. DEL-1 interacts with several integrins via its RGD motif or other structural features, modulating cell adhesion, angiogenesis, inflammation, and bone formation. It is considered a promising therapeutic target for chronic inflammatory diseases, cardiovascular/metabolic disorders, ischemic injury recovery strategies, and autoimmune conditions.

Mechanism of action

DEL-1 interacts with integrins (αvβ3, αvβ5, αLβ2, αMβ2) and other molecules to modulate cell adhesion, migration, angiogenesis, immune cell recruitment, and efferocytosis, impacting inflammatory and vascular processes. It also affects osteogenic differentiation via integrin signaling.
