Diacylglycerol kinase (DGK or DAGK) is a family of enzymes that catalyzes the ATP-dependent phosphorylation of diacylglycerol (DAG) to phosphatidic acid (PA). This reaction plays a crucial role in cellular signaling by regulating the levels of two important bioactive lipids: DAG and PA, both of which act as second messengers in various signal transduction pathways. DGKs serve as molecular switches, terminating DAG-mediated signaling while simultaneously activating PA-mediated pathways. There are at least ten known mammalian isoforms of DGK with tissue-specific expression patterns and regulatory roles. Targeted modulation of specific DGK isoforms is being explored for diseases involving aberrant lipid signaling—including neuronal disorders and cancer immunotherapy applications.
