Diacylglycerol kinase delta is a cytoplasmic enzyme encoded by the DGKD gene in humans. It catalyzes the ATP-dependent phosphorylation of diacylglycerol (DAG) to form phosphatidic acid (PA), thereby regulating the relative levels of these two important lipid second messengers. Both DAG and PA are involved in various signaling pathways, affecting cellular processes such as proliferation, differentiation, and survival. By modulating DAG and PA levels, DGKD plays an important role in signal transduction and cellular homeostasis. The enzyme contains functional domains, including a kinase domain and, in some isoforms, a sterile alpha motif (SAM) domain that mediates protein-protein interactions. Variants of the DGKD gene arise from alternative splicing, resulting in at least two distinct isoforms in humans[1][3][5][6][7].
