Overview

Diacylglycerol kinase iota is an enzyme that phosphorylates the lipid messenger diacylglycerol (DAG) to produce phosphatidic acid (PA), regulating the respective levels of these two important bioactive lipids[2][3][5][6][7]. This activity is central to lipid metabolism and signal transduction in eukaryotic cells[1][2][3][6]. DGKI belongs to a conserved family of DGKs characterized by a catalytic domain and multiple cysteine-rich (C1) domains[1][5]. Tissue expression studies show particularly high levels in the brain and retina, suggesting a role in the central nervous system and visual processing[4][5]. Dysregulation of diacylglycerol kinase enzymes, including DGKI, has been connected to cancer and metabolic diseases[1]. The human protein is encoded by the DGKI gene.
