Diacylglycerol kinase theta (DGKQ) is an enzyme that phosphorylates diacylglycerol (DAG) to generate phosphatidic acid (PA), acting as a central regulatory switch between signaling pathways mediated by these two important lipid second messengers. The protein contains cysteine-rich domains, a proline-rich region, and a pleckstrin homology domain with an overlapping Ras-associating domain, and is localized to the nuclear speckle domains. DGKQ is implicated in various biological processes, including the regulation of the phosphatidylinositol cycle during cell signal transduction, synaptic vesicle endocytosis, and steroidogenic gene transcription, with documented disease associations such as retinal degeneration and postural kyphosis[1][2][3][7].
