Diacylglycerol O-acyltransferase 1 (DGAT1) is a key enzyme that catalyzes the final step in triacylglycerol (TAG) synthesis: the conversion of diacylglycerol (DAG) and fatty acyl-CoA to triacylglycerol. It is essential for fat storage, energy metabolism, and lipid homeostasis. Loss-of-function mutations are associated with congenital diarrheal disorders due to defective fat absorption. DGAT1 inhibitors are being explored as therapeutic agents for treating obesity-related conditions and other metabolic disorders.
