Overview

DIAPH2 antisense RNA 1 (DIAPH2-AS1) is a long non-coding RNA gene expressed in humans, located on the X chromosome. It does not encode a protein. Recent research indicates DIAPH2-AS1 acts as an oncogenic lncRNA in gastric cancer, particularly promoting neural invasion, cell migration, and invasion. Mechanistically, DIAPH2-AS1 interacts with the RNA-binding protein NSUN2, stabilizing it by preventing ubiquitin-proteasome-mediated degradation. NSUN2 in turn enhances the stability of NTN1 mRNA via m5C modification, promoting tumor progression. DIAPH2-AS1 is also reported to function as an early signal in T-cell activation, suggesting a role in immune response. There are no known approved drugs that directly target DIAPH2-AS1 itself, and it is not classified as a receptor, enzyme, or transporter, but rather as a regulatory non-coding RNA of emerging clinical interest in oncology and immunology[1][3][5].
