DIAPH3 antisense RNA 1 is a long noncoding RNA transcribed from the DIAPH3 gene locus in the antisense direction. It is thought to regulate its putative target, DIAPH3—a protein-coding gene involved in actin cytoskeleton regulation and cancer metastasis—though the exact function of DIAPH3-AS1 itself remains to be fully elucidated. Expression studies demonstrate DIAPH3-AS1 is differentially expressed in cancer tissues and is genomically located near RB1, a known tumor suppressor. Association studies have identified DIAPH3-AS1 as a possible biomarker for more aggressive or metastatic forms of cancer, especially triple-negative breast cancer, and have correlated its expression with worse patient outcomes. No drugs directly target DIAPH3-AS1 to date, but its emerging disease associations suggest further research into its therapeutic and prognostic potential is warranted.
Not established for DIAPH3-AS1, as no drugs or experimental therapeutics are known to act on it directly. Reported mechanistic hypotheses focus on its role in regulation of adjacent DIAPH3 gene expression and possible influence on oncogenic signaling pathways.
