Overview

DIAPH3 antisense RNA 1 is a long noncoding RNA transcribed from the DIAPH3 gene locus in the antisense direction. It is thought to regulate its putative target, DIAPH3—a protein-coding gene involved in actin cytoskeleton regulation and cancer metastasis—though the exact function of DIAPH3-AS1 itself remains to be fully elucidated. Expression studies demonstrate DIAPH3-AS1 is differentially expressed in cancer tissues and is genomically located near RB1, a known tumor suppressor. Association studies have identified DIAPH3-AS1 as a possible biomarker for more aggressive or metastatic forms of cancer, especially triple-negative breast cancer, and have correlated its expression with worse patient outcomes. No drugs directly target DIAPH3-AS1 to date, but its emerging disease associations suggest further research into its therapeutic and prognostic potential is warranted.
