Dickkopf-related protein 1 (DKK1) is a secreted glycoprotein that acts as an antagonist of the Wnt/β-catenin signaling pathway. It inhibits Wnt signaling by binding to LRP5/6 co-receptors, preventing downstream activation. DKK1 plays critical roles in embryonic development, bone homeostasis, and is implicated in various diseases including cancer, bone disorders, and cardiovascular disease. It is being investigated as a therapeutic target, particularly in oncology and immunotherapy, with anti-DKK1 antibodies showing promise in preclinical studies.
DKK1 binds to LRP5/6 co-receptors, preventing activation of downstream Wnt signaling events. Anti-DKK1 antibodies neutralize DKK1, promoting Wnt signaling and potentially reducing immune suppression.
