Overview

Dietary phospholipids, primarily consisting of phosphatidylcholine, are critical components of the gastrointestinal (GI) environment that serve both protective and digestive functions. They form a hydrophobic, surfactant-like layer on the surface of the mucus gel, which acts as a primary biophysical barrier against luminal aggressors such as gastric acid, bile salts, and digestive enzymes (Lichtenberger et al., 1995). In addition to barrier maintenance, these phospholipids are essential for the emulsification of dietary fats, facilitating their breakdown by pancreatic lipases and subsequent absorption in the small intestine (Dial et al., 2008). Clinical research has highlighted the importance of these molecules in inflammatory bowel diseases; for instance, patients with ulcerative colitis often exhibit a significant depletion of phosphatidylcholine in their colonic mucus, which correlates with increased disease activity (Ehehalt et al., 2010). Therapeutic strategies like delayed-release phosphatidylcholine (e.g., LT-02) are designed to replenish this lipid layer and restore mucosal integrity. Furthermore, the interaction between phospholipids and non-steroidal anti-inflammatory drugs (NSAIDs) is pharmacologically significant, as NSAIDs can disrupt this protective layer, leading to mucosal injury and ulceration (Lichtenberger et al., 2006).
