Overview

Dietary starches and polysaccharides are complex carbohydrates consisting of glucose monomers linked by glycosidic bonds, serving as a major energy source in the human diet. They are primarily categorized into amylose, amylopectin, and resistant starches, which vary in their digestibility and physiological impact (StatPearls, 2023). In the digestive tract, these molecules are hydrolyzed by salivary and pancreatic alpha-amylase and subsequently by brush-border enzymes like alpha-glucosidase into absorbable monosaccharides (PubMed, 2021). While these polysaccharides are not therapeutic targets in the traditional sense, they are the substrates for enzymes that are key targets in the management of metabolic diseases. For instance, alpha-glucosidase inhibitors like acarbose and miglitol competitively inhibit the breakdown of these starches to prevent rapid spikes in postprandial blood glucose levels in type 2 diabetes patients (PubChem, 2024). Furthermore, non-digestible polysaccharides, known as dietary fiber, play a significant role in cardiovascular health and weight management by modulating gastric emptying and gut hormone signaling (NIH, 2022). These substances also serve as critical substrates for the gut microbiota, where their fermentation produces short-chain fatty acids that influence systemic inflammation and metabolic homeostasis (Nature, 2020).
