Dietary triglycerides and other dietary fats are the primary form of lipid energy consumed by humans, consisting of a glycerol backbone esterified with three fatty acids (StatPearls, 2023). These molecules are essential for the absorption of fat-soluble vitamins (A, D, E, and K) and serve as critical components of cellular membranes and signaling pathways (NIH, 2022). However, chronic excessive intake is strongly linked to the development of obesity, type 2 diabetes, and cardiovascular diseases (Mayo Clinic, 2023). In a pharmacological context, these fats are not targets in the traditional sense of a protein receptor; rather, they are the substrates whose processing is modulated (PubChem, 2024). Drugs like orlistat inhibit the enzymes (lipases) responsible for their breakdown, while others like ezetimibe block the transport of specific dietary fats like cholesterol across the intestinal lumen, effectively reducing the systemic burden of these lipids (FDA, 2021).
Inhibition of gastrointestinal lipases (e.g., pancreatic lipase) to prevent the hydrolysis of triglycerides into absorbable free fatty acids and monoglycerides (StatPearls, 2023); inhibition of the NPC1L1 transporter to prevent cholesterol absorption (PubMed, 2022); or sequestration of bile acids to impair lipid emulsification and absorption (NIH, 2023).
