Diethylenetriaminepentaacetic acid (DTPA), also known as pentetic acid, is a synthetic polyamino carboxylic acid that acts as a potent chelating agent rather than a conventional biological target such as a receptor or enzyme. It possesses a high affinity for polyvalent metal ions, including transuranic elements like plutonium, americium, and curium, as well as lanthanides like gadolinium (PubChem CID 3053). In clinical practice, DTPA is utilized primarily for decorporation therapy to enhance the renal clearance of radioactive contaminants following internal exposure (FDA, Ca-DTPA/Zn-DTPA Labels). Beyond toxicology, DTPA serves as a critical carrier molecule in diagnostic medicine; when complexed with radioisotopes or paramagnetic metals, it facilitates specialized imaging procedures such as renal scintigraphy and contrast-enhanced MRI (StatPearls, Pentetic Acid). Because it can non-selectively bind essential endogenous metals, its therapeutic administration requires careful monitoring of mineral status and kidney function.
DTPA functions as a polydentate ligand that sequesters polyvalent metal ions by forming stable, water-soluble coordination complexes (chelates). These complexes are biologically inert and are rapidly excreted from the body via glomerular filtration in the kidneys.
