Overview

Diethylenetriaminepentaacetic acid (DTPA) chelate haptens are synthetic chemical moieties used as essential components in pre-targeted radioimmunotherapy (PRIT) and diagnostic nuclear medicine (PubChem CID 3053). DTPA is a potent aminopolycarboxylic acid chelator that forms highly stable complexes with various metallic radionuclides, such as Indium-111, Yttrium-90, and Gadolinium (StatPearls, "Pentetic Acid"). In the context of pre-targeting, these metal-DTPA complexes serve as haptens—small molecules that are specifically recognized by the capture arm of a bispecific antibody (Goldenberg et al., J Nucl Med, 2006). The clinical procedure involves first administering a bispecific antibody that localizes to a tumor-associated antigen, followed by the administration of the radiolabeled DTPA hapten (Le Doussal et al., J Nucl Med, 1990). This strategy allows the radioactivity to rapidly bind to the pre-localized antibody at the tumor site while the unbound fraction is quickly cleared through the kidneys, significantly reducing systemic radiation toxicity compared to directly labeled antibodies (PubMed, PMID 2107436). Additionally, DTPA is used in its salt forms as a decorporation agent to treat internal contamination with transuranic elements (FDA, "Ca-DTPA and Zn-DTPA Information").
