Diethylenetriaminepentaacetic acid-iodine-131 hapten (di-DTPA-¹³¹I hapten) is a synthetic, radiolabeled, low-molecular-weight compound consisting of diethylenetriaminepentaacetic acid (DTPA) chelator conjugated to radioactive iodine-131. It is designed to function as a hapten—an antigenic determinant too small to generate an immune response by itself but able to be recognized by anti-hapten antibodies. In pretargeted radioimmunotherapy protocols, a bispecific antibody is first administered to bind a tumor-associated antigen and to recognize the hapten; the di-DTPA-¹³¹I hapten is then administered, which is selectively delivered and retained at the tumor site via the antibody, allowing targeted delivery of radioactivity for imaging or therapy. Di-DTPA-¹³¹I haptens are not natural biological targets, do not have endogenous ligands, and only function as components in engineered therapeutic or diagnostic platforms.
It is not a pharmacological target but acts as an antigenic determinant for antibody recognition; its mechanism of biological utility is being recognized by a specific antibody.
