Differentially expressed in FDCP 6 homolog (DEF6) is a **guanine nucleotide exchange factor (GEF)** that promotes the exchange of GDP for GTP on small G proteins, regulating their activation state[1][2]. DEF6 is particularly expressed in immune cells, where it functions as a signaling adaptor protein, crucially orchestrating pathways involved in T cell activation, cytoskeletal rearrangements, and immune synapse formation. As a GEF, DEF6 controls the activity of several Rho GTPases, thereby integrating signals that shape cell morphology, motility, and immune responses. Dysregulation of DEF6 is implicated in immune-mediated diseases, inflammation, and some cancers, with altered DEF6 expression linked to abnormal lymphocyte activation or malignancies. DEF6 is not yet a broadly established drug target, and specific pharmacological modulators or biomarkers are currently not established or FDA-approved.
