Overview

Dihydroceramide desaturase 1 (DEGS1) is a key enzyme in sphingolipid metabolism, catalyzing the desaturation of dihydroceramide to ceramide through introduction of a 4,5-trans double bond. This reaction constitutes the final step in the de novo ceramide biosynthesis pathway, which occurs in the endoplasmic reticulum. DEGS1 activity is essential for proper cellular function, as ceramides serve as central signaling molecules regulating apoptosis, cell growth, autophagy, and stress responses. Disruption of DEGS1, by genetic deletion or chemical inhibition, alters the balance of bioactive sphingolipids and is linked to cancer progression, neurodegeneration, and metabolic dysfunction. Modulation of DEGS1 is being explored therapeutically in cancer and metabolic disease, though safety challenges remain due to ceramide pathway's fundamental role in cell physiology.
