Overview

Dihydrofolate reductase pseudogene 3 (DHFRP3, hDHFR-psi 3) is a processed, intronless pseudogene associated with the DHFR gene family. Pseudogenes such as DHFRP3 arise through genomic duplication and subsequent loss of protein-coding potential. There are multiple DHFR-related pseudogenes in the human genome, commonly labeled as DHFRP1, DHFRP2, DHFRP3, and DHFRP4 (sometimes also referred to as DHFRL1)[6][7]. Unlike the functional DHFR enzyme, which is essential for folate metabolism and is a key drug target (especially in oncology and anti-infective therapy), DHFRP3 does not encode an active protein and is not implicated in any biological process, disease role, or drug response[2][6][7]. There is no evidence that DHFRP3 is expressed or encodes a functional protein, nor is there any indication that it serves as a direct therapeutic target, biomarker, or interacts with drugs. Only pseudogene DHFRL1 has been shown to have some expression and partial enzyme activity; the other pseudogenes, including DHFRP3, do not[7].
