Dihydrofolate synthetase (DHFS) is an enzyme that catalyzes the ATP-dependent ligation of L-glutamate to 7,8-dihydropteroate, forming 7,8-dihydropteroylglutamate (dihydrofolate). This enzyme is a key component of the folic acid biosynthesis pathway in bacteria and is absent in mammals, making it a validated target for antimicrobial drug discovery. Inhibition of DHFS disrupts folate synthesis, which is essential for nucleotide synthesis and amino acid metabolism, leading to bacterial cell death.
