Overview

Dihydropteroate synthase (DHPS) is an enzyme that catalyzes the condensation of 6-hydroxymethyl-7,8-dihydropterin pyrophosphate (DHPPP) with para-aminobenzoic acid (pABA) to produce 7,8-dihydropteroate, a precursor for folic acid synthesis. It is a key enzyme in the folate biosynthesis pathway of bacteria, lower eukaryotes, and plants. DHPS is the target of sulfonamide antibiotics, which act as competitive inhibitors of pABA binding. Resistance to sulfonamides has emerged due to mutations affecting the pABA binding site on DHPS.
