Dihydropyrimidinase-related protein 3 (DPYSL3) is a cytoskeletal regulatory protein belonging to the collapsin response mediator protein family, often referred to as CRMP4. It participates in axon guidance, migration, and cytoskeleton remodeling in nervous system development and has critical roles in cell cycle regulation and cell motility. DPYSL3 interacts with actin and tubulin, controlling microtubule polymerization and stabilization, and influences processes such as epithelial-to-mesenchymal transition (EMT). In cancer biology, DPYSL3 serves as a biomarker and modulator of motility and metastasis, having context-dependent roles—acting as a metastasis suppressor or promoter in different tumor types. Its activity is tightly regulated by phosphorylation, impacting its interactions with other kinases (especially PAK family members) and cytoskeletal proteins. DPYSL3 is involved in diseases including several cancers, neurodevelopmental disorders, and epilepsy[1][2][3][4].
