Dihydropyrimidinase-related protein 4 (DPYSL4) is a member of the collapsin response mediator protein (CRMP) family that plays a critical role in neuronal development, especially in neurite outgrowth, dendrite formation, and axon guidance by mediating cytoskeletal remodeling in response to class 3 semaphorins[1][5]. Beyond the nervous system, DPYSL4 is a direct transcriptional target of the tumor suppressor p53, and has been shown to regulate mitochondrial function, oxidate phosphorylation, ATP production, and cellular energy metabolism in both normal and cancer cells[3]. DPYSL4 functions as a tumor suppressor by suppressing tumor growth and metastasis through energy modulation. Its expression is reduced in several cancer types, with lower levels correlated to worse prognosis in breast and ovarian cancer[3]. Epigenetic changes of DPYSL4 have also been noted in hepatocellular carcinoma, serving as a potential biomarker for cancer detection[1]. DPYSL4 has not been reported as a direct drug target, and there are currently no known drugs or inhibitors acting specifically on this protein.
