Overview

DIM1 rRNA methyltransferase and ribosome maturation factor (DIMT1) is an enzyme essential for the biogenesis of the small ribosomal subunit in eukaryotes[1][2][3]. It specifically dimethylates two adjacent adenosines near the decoding site of 18S rRNA, a critical step required for proper pre-rRNA processing and ribosome assembly[1][2][3][5]. This function is conserved from yeast (Dim1) to humans (DIMT1/DIMT1L), but in humans, the modification occurs in the nucleus[1][2][3]. The presence of DIMT1 on ribosomal precursors, rather than its catalytic activity per se, is essential for downstream steps in ribosome maturation, positioning it as a key quality control factor in the maturation process[2][3]. Diseases associated with DIMT1 include Diamond-Blackfan anemia and localized osteosarcoma[1], and its dysregulation may play a role in cancer and other ribosome-related disorders[1]. No small-molecule drugs or clinical inhibitors specifically targeting DIMT1 are currently known.
