Overview

DIO3 opposite strand upstream RNA (DIO3OS) is a long non-coding RNA located on human chromosome 14, transcribed opposite and overlapping with the DIO3 (deiodinase, iodothyronine, type III) gene. DIO3OS is not translated into protein but is thought to have a regulatory function in the imprinting and monoallelic expression of the DIO3 gene, particularly during fetal development. DIO3OS is expressed in several tissues, including gut, liver, lung, placenta, and yolk sac in mouse, with human orthologous expression expected to be similar[3]. Diseases including thyroid cancer and pancreatic cancer have been associated with DIO3OS, although its direct role in disease is still under investigation[2]. DIO3OS is not considered a therapeutic target as it is not an enzyme, receptor, transporter, or protein-coding gene, but rather functions in gene regulation as a long non-coding RNA[2][6].
