Overview

Dipeptidyl peptidase 9 (DPP9) is a serine protease enzyme that cleaves Xaa-Pro dipeptides from the N-termini of proteins. It plays a role in regulating immunity, energy metabolism, cell survival, proliferation, apoptosis, and adhesion/migration. Unlike DPP4, DPP9 is primarily located in the cytoplasm and nucleus. Altered DPP9 function is linked to type 2 diabetes, obesity, cancer, and immune disorders. It is a key inhibitor of caspase-1-dependent pyroptosis in monocytes/macrophages and interacts with inflammasome regulators like NLRP1 and CARD8. It also interacts with Filamin A and Syk kinase, modulating B-cell signaling.
