Overview

Diphosphoinositol polyphosphate phosphohydrolase NUDT4B is a cytosolic enzyme that cleaves β-phosphate from diphosphate groups in diphosphoinositol polyphosphates, such as PP-InsP5, PP-InsP4, and bisdiphosphoinositol tetrakisphosphate. The enzyme contributes to the turnover of these high-energy molecules, which act as molecular switches in intracellular signaling processes and possibly in regulating intracellular trafficking. It can also catalyze the hydrolysis of dinucleoside oligophosphate (Ap6A), forming ADP and p4a, and can hydrolyze 5-phosphoribose 1-diphosphate. NUDT4B is closely related to NUDT4 and has several transcript variants; however, its full clinical significance and therapeutic potential remain under-investigated. - NUDT4B is distinct from the closely related NUDT4 gene/protein, and from NUDT4P1, which is a pseudogene. - Its classification is well-supported as an enzyme in the Nudix hydrolase family, but direct links to disease or clinical applications are limited. - Some sources list "Diphosphoinositol polyphosphate phosphohydrolase 2" as an alternative name, but genomic and protein databases confirm that NUDT4B is the specific gene/protein indicated.
