CRM197 is a genetically detoxified, non-toxic mutant of diphtheria toxin used as a carrier protein in conjugate vaccines to enhance the immunogenicity of polysaccharides and haptens. It is produced by a single amino acid substitution (G52E) that abolishes ADP-ribosyltransferase activity. It acts by converting T-cell independent antigens into T-cell dependent ones. CRM197 has a well-established safety profile and is included in several globally used pediatric vaccines.
