Overview

CRM197 is a non-toxic mutant of the diphtheria toxin, widely used as a carrier protein in conjugate vaccines to enhance immunogenicity of polysaccharides and haptens. It is produced by a single amino acid substitution (Gly52→Glu), abolishing ADP-ribosylation activity and rendering it non-toxic, while retaining immunological identity to wild-type diphtheria toxin. Used in vaccines against pneumococcal, meningococcal, and Haemophilus influenzae type b diseases.
