Overview

A diphtheria toxin-derived T-helper epitope is a short peptide sequence derived from the diphtheria toxin (DT), specifically designed to be recognized by CD4+ T-helper cells. These epitopes are commonly used as immunological adjuvants or carrier sequences in vaccine design to enhance immune responses against weakly immunogenic antigens. The most widely referenced epitope is DT(331–345), corresponding to amino acids 331 through 345 of the diphtheria toxin protein. It acts as a "universal" helper epitope, stimulating human CD4+ T cells across diverse HLA backgrounds. When covalently linked to poorly immunogenic antigens, it enhances both humoral and cellular immune responses. Applications include cancer vaccines, allergy vaccines, and general immunogenicity enhancement.
