DIRAS family GTPase 2 (DIRAS2) is a small monomeric GTP-binding protein of the Ras superfamily, characterized by low intrinsic GTPase activity and predominantly present in its GTP-bound form[2][7][6]. Its protein product is believed to participate in signal transduction pathways, analogous to other Ras superfamily members, which regulate diverse cellular processes such as proliferation and differentiation[1][2]. DIRAS2 is expressed mainly in brain regions including the cerebral cortex, occipital and frontal lobes, and cerebellum, implicating a possible role in neurological activity[1][3]. Genetic studies have linked promoter variants in DIRAS2 to susceptibility for neurodevelopmental disorders, particularly attention-deficit/hyperactivity disorder (ADHD), where altered expression may impact prefrontal brain activity related to response inhibition[1]. The precise physiological role of DIRAS2 remains largely uncharacterized, and currently, it is not recognized as a direct therapeutic target nor are any drugs known to act upon it[2][7][1].
