Discoidin domain-containing receptor 1 (DDR1) mRNA is the transcript encoding the DDR1 protein, a unique receptor tyrosine kinase that is activated by various types of collagen rather than soluble growth factors (UniProt P35908). DDR1 plays a critical role in mediating cell-extracellular matrix interactions, influencing processes such as cell adhesion, proliferation, and remodeling of the matrix (NCBI Gene 780). In pathological states, DDR1 mRNA is frequently overexpressed, contributing to the progression of fibrotic diseases in the lungs and kidneys, as well as promoting tumor invasion and metastasis in several cancers (PubMed 25650995). Targeting DDR1 at the mRNA level using technologies like small interfering RNA (siRNA) or antisense oligonucleotides (ASOs) offers a strategy to silence the expression of the protein entirely, thereby inhibiting both its catalytic and non-catalytic signaling functions. This approach is particularly relevant in conditions where small molecule kinase inhibitors may not fully block the protein's scaffolding roles or where high specificity is required to avoid off-target effects on related kinases (PubMed 30254247).
