Discoidin Domain Receptor Tyrosine Kinase 2 (DDR2) is a receptor tyrosine kinase activated by fibrillar collagens. It plays a critical role in cell growth, differentiation, migration, and extracellular matrix remodeling. Dysregulation of DDR2 is implicated in diseases such as fibrosis, arthritis, and various cancers.

Mechanism of action

DDR2 is activated upon binding to specific types of fibrillar collagens—primarily types I, III, and X—via its extracellular discoidin domain. This interaction triggers autophosphorylation within the cytoplasmic kinase domain and initiates downstream signaling cascades that regulate cell behavior and ECM remodeling. Inhibition of DDR2 kinase activity or collagen binding is a potential therapeutic strategy.
