Discs large MAGUK scaffold protein 1 (DLG1) is a modular scaffolding protein and a member of the membrane-associated guanylate kinase (MAGUK) family.[2][1] It contains multiple interaction domains including L27, PDZ, SH3, and a catalytically inactive guanylate kinase domain.[2] DLG1 is evolutionarily conserved and plays a central role in establishing and maintaining epithelial cell polarity, forming the Scribble polarity complex at basolateral membranes. It organizes and anchors diverse receptors, ion channels, and cytoskeletal elements, most notably in neuronal synapses (clustering glutamate and potassium channels) and at cell-cell junctions.[2][1][3] DLG1 regulates vesicular trafficking for ciliary and membrane proteins in kidney epithelial cells, with dysfunction resulting in disorders like CAKUT, cystic kidneys, and developmental abnormalities. It is not an enzyme or receptor directly targeted by drugs but serves critical scaffold and signaling roles in tissue organization and function.[2][1][3]
