Disintegrin and metalloproteinase domain-containing protein 21 (ADAM21) is a member of the ADAM (A Disintegrin And Metalloproteinase) family, a group of membrane-anchored proteins with diverse roles, including regulation of cell adhesion, migration, proteolysis, and signaling[1][2][3][4][5]. ADAM21 is predominantly expressed in the testis, especially in Leydig and Sertoli cells, and is thought to be involved in sperm maturation and/or fertilization[4][5]. However, knockout mouse studies indicate that ADAM21 is dispensable for male fertility and spermatogenesis in mice[4]. ADAM21 has not been identified as a functional or established therapeutic target, and there are no approved drugs known to interact with it. Its precise biological roles in humans remain incompletely defined.
