Overview

Disintegrin and metalloproteinase domain-containing protein 33 (ADAM33) is a type I transmembrane zinc-dependent metalloprotease and a member of the ADAM family, structurally related to snake venom disintegrins. Primarily expressed in mesenchymal cells including airway fibroblasts, myofibroblasts, and smooth muscle cells, ADAM33 has a structure featuring pro-metalloprotease-like, disintegrin-like, cysteine-rich, EGF-like, and cytoplasmic domains. Genetic studies have identified ADAM33 as the first asthma-susceptibility gene linked to bronchial hyperresponsiveness, with polymorphisms associated with asthma in many populations. Its biological functions include modulating cell adhesion, proliferation, differentiation, and promoting angiogenesis. ADAM33 is strongly implicated in airway remodeling in asthma, acting at the interface of structural and inflammatory pathways, though its precise function and mechanisms in disease are still being investigated.
