Disks large-associated protein 1 (DLGAP1) is a major postsynaptic scaffold protein highly enriched in the brain’s synaptosomal fraction and localized at the postsynaptic density (PSD) of glutamatergic neurons. DLGAP1 links core synaptic scaffolds such as the DLG/PSD-95 and SHANK families, facilitating the assembly of protein supercomplexes that organize synaptic signaling and plasticity. Genetic variants in DLGAP1 are associated with schizophrenia, autism spectrum disorder, and obsessive-compulsive disorder, with evidence suggesting that loss of function impairs PSD organization and sociability in animal models. DLGAP1's role is structural and regulatory within the synapse, forming connections between scaffolding proteins but not acting as a receptor or enzyme. No known drugs currently target DLGAP1 directly; rather, its dysfunction contributes to risk of complex brain diseases.
