Overview

Disks large-associated protein 2 (DLGAP2) is a membrane-associated scaffold protein localized at the postsynaptic density of excitatory synapses in neuronal cells, where it plays a key organizational role[3][4][5]. DLGAP2 connects neurotransmitter receptors such as NMDA and AMPA receptors to cytoskeletal and signaling proteins through interactions with other scaffold proteins like PSD-95/SAP90 (DLG4), SHANK, and Homer[3][4][6]. By contributing to the physical and functional architecture of the postsynaptic density, DLGAP2 participates in regulating synaptic scaling—a core process in neuronal signaling and plasticity. Genetic and functional studies link DLGAP2 to a range of neurological and neurodevelopmental disorders, particularly autism spectrum disorder, epilepsy, and schizophrenia[3][4][5]. There are currently no known drugs that target DLGAP2 directly for therapeutic use or with established mechanisms of drug interaction.
