Overview

Disks large-associated protein 3 (DLGAP3; SAPAP3) is a neuronal scaffold/adaptor protein encoded by the DLGAP3 gene[1][2]. It is a key structural component of the postsynaptic density (PSD) in excitatory synapses, facilitating the organization and assembly of protein complexes at the synapse[1][3]. DLGAP3 interacts with postsynaptic proteins including PSD-95/SAP90 (DLG4), SHANK family scaffold proteins, and links ionotropic (NMDA, AMPA) and metabotropic glutamate receptors (mGluR) to the subsynaptic cytoskeleton[3]. It regulates synaptic scaling and homeostatic plasticity by controlling receptor turnover and postsynaptic signaling dynamics[3]. Genetic variation or loss of DLGAP3 disrupts synaptic function and is associated with neuropsychiatric conditions such as trichotillomania, body dysmorphic disorder, and may contribute to disease mechanisms in schizophrenia, autism spectrum disorders, and other brain disorders[1][3]. DLGAP3 is not an established therapeutic target; it lacks direct drug modulators and is not used as a biomarker in clinical practice. Its disruption or dysregulation may be relevant to behavioral symptoms and synaptic pathophysiology in neuropsychiatric disease[3].
