Overview

Disks large-associated protein 4 (DLGAP4) is a synaptic scaffold protein found at the postsynaptic density of neurons, where it interacts with signaling molecules, potassium channels, and major synapse organizers such as PSD-95[3]. It is a member of the membrane-associated guanylate kinase (MAGUK) family and plays vital roles in the molecular organization of synapses, neuronal signal transduction, and regulation of the actin cytoskeleton[1][2]. DLGAP4 is essential for proper neuronal migration and cortical development, interacting with proteins involved in adherens junctions and actin dynamics[1]. Mutations in DLGAP4 are associated with neurological disorders including epilepsy, intellectual disability, schizophrenia, and others, and aberrant expression is linked to tumor progression and prognosis in hepatocellular carcinoma[1][4]. No approved drugs are known to directly target DLGAP4, but it may serve as a biomarker in some disease contexts[4].
