Overview

Disks large MAGUK scaffold protein 4 (DLG4), also known as postsynaptic density protein 95 (PSD-95), is a major synaptic scaffolding protein in the membrane-associated guanylate kinase (MAGUK) family. It is highly enriched at postsynaptic densities of neurons and is essential for synaptic organization, clustering ionotropic glutamate receptors (NMDA, AMPA) and potassium channels, and regulating their trafficking, anchoring, and signaling. DLG4/PSD-95 underlies synaptic plasticity, learning, and memory, and is implicated in several neurological and developmental disorders due to its central role in mediating synaptic protein interactions and maintaining excitatory-inhibitory balance in the brain

Mechanism of action

Not direct; PSD-95/Disks large MAGUK scaffold protein 4 acts as a synaptic scaffold and is targeted experimentally by peptide inhibitors to disrupt protein-protein interactions involving NMDA receptors, which may modulate excitotoxicity and plasticity in CNS disease models
