Overview

Disrupted in renal carcinoma 1 (DIRC1), also known as Disrupted in renal cancer protein 1, is a long non-coding RNA gene originally found disrupted in families with inherited renal cell carcinoma due to a chromosomal translocation t(2;3)(q33;q21). DIRC1 does not encode a functional protein; rather, it acts as an RNA regulator. Its aberrant expression is associated with cancer susceptibility and progression, including familial renal cell carcinoma and stomach adenocarcinoma, where high DIRC1 expression correlates with worse prognosis, more advanced tumor stage, and increased immune cell infiltration. DIRC1 is considered a novel prognostic biomarker for several cancers, but there are no drugs or therapies directly targeting it, and its precise molecular mechanisms remain incompletely understood.
