Distal-less homeobox 2 (DLX2) is a homeobox transcription factor that plays essential roles in vertebrate development, notably in the formation of the forebrain, craniofacial structures, and specification of neuronal subtypes[1][5][7]. As a member of the DLX gene family (DLX1–DLX6), DLX2 is involved in the differentiation of GABAergic interneurons from neural progenitors in the subventricular zone, the regulation of cranial neural crest cell development, and the directed osteogenic differentiation of mesenchymal stem cells[2][3][5][7]. DLX2 functions mainly as a transcriptional activator, sometimes partnering with other transcription factors such as PAX6, and is regulated by signaling pathways including BMP (bone morphogenetic protein). Mutations in DLX2 are associated with congenital disorders affecting limb and craniofacial formation[7]. There are no documented small molecule drugs, approved therapeutics, or direct modulators of DLX2 in current clinical use.
